We See Something!!!!

One path of the microscope (astigmatic path) has been assembled and we were successful in looking at 200 nm red fluorescent beads. Currently we are in the process of the running tests on the microscope so that we can adjust components if needed. The stage is not as stable as we need it to be. We are currently talking to the vendor (Smaract) regarding this issue. The stage drifts ~ 120 nm over 30 mins which is not ideal for localization microscopy. To test the resolution of the system, we measured the PSF.

The Full-width half maximum (FWHM) was measured to be ~318.53 nm. The fluorescent bead was measured using a 1.4 NA oil objective.
